ترادف نوکلئوتیدی ژن p25 جدایه لرستان ویروس Beet necrotic yellow vein virus

در این مطالعه نمونه­های چغندرقند مبتلا به ریشه ریشی از
استان لرستان جمع­آوری، پس از تایید آلودگی به روش الیزا، استخراج RNA و ترانویسی معکوس با استفاده از آغازگرهای اختصاصی،
ژن p25 در آنها تکثیر و پس از خالص سازی تعیین ترادف گردید. با
آنالیز ژن p25 جدایه­ی لرستان و بررسی آمینو­اسید­های
موقعیت 67-70 برای اولین بار تیپ این ویروس در
استان لرستان A (BNYVV type A)
گزارش
می­گردد. رسم درخت تبارزایی بر پایه­­ی ژن p25 مشخص کرد که این جدایه رابطه نزدیکی با جدایه­ی همدان داشته و
این
گروه به عنوان گروه خواهری جدایه­ی آلمان محسوب می­شود.

Rhizomania is a devastating disease of sugarbeet caused
by Beet necrotic yellow vein virus (BNYVV) from the genus Benyvirus. It
is considered as the most important sugarbeet disease in Iran. The genome of
BNYVV is composed of five RNA molecules with specific functions. The virus has
three types of A, B and P. All three have been reported from Iran, but the type
of virus in Lorestan province has remained undetermined. In this study, sugarbeet
plant showing rhizomania symptoms were sampled from Lorestan and the presence
of BNYVV in the samples was confirmed by ELISA. PCR amplification of p25 gene with
specific primers followed by sequencing and analysis were used to determine the
type of BNYVV in the Lorestan province. We report presence of type A of the
virus based on p25 nucleotide sequence and identity of amino acids in the 67-70
position. Phylogenetic tree constructed based on nucleotide sequence of the p25
gene showed that this isolate is closely related to isolates of Hamedan and the
group is a sister group of German isolates.
